Peptide Microarray

Peptide microarrays offer great potential as a screening tool for peptidomics experiments and have been widely used in fields ranging from immunological research to systems biology. Peptides are also short chains (primary structure) composed of amino acids, but their size (less than 100 amino acids) is usually much smaller than that of proteins with higher structures. Peptide microarrays have become a promising technique for simultaneous high-throughput analysis of peptide characteristics by allowing the parallel analysis of thousands of peptides in one run. The synthesized peptides are spotted in a grid layout on a glass slide, which allows screening of thousands of peptides in a single experiment with only a small amount of sample.

Compared with microarrays formed from full-length proteins, peptide microarrays have the advantage of requiring only a small amount of peptides. This means that the production method of chemically synthesized peptides in microarrays is more economical and more effective than proteins purified from biological sources, and peptide microarrays are easier to store than protein microarrays. In addition, compared with larger proteins, peptides have a high degree of specificity, usually a single epitope. Compared with protein microarrays, peptide microarrays are more suitable for the following applications. Peptides can recognize specific sites of protein-protein interactions and can recognize discrete protein structures of drug targets (such as enzyme binding sites). In addition, peptides can also be used to generate highly specific antibodies against discrete epitopes, which can distinguish similar or isotype proteins.

Applications of Peptide Microarrays

Peptide microarrays have been widely used in biomarker discovery, immune monitoring, antibody epitope mapping, multiple epitope mapping, or used to detect and verify protein interactions. Applications range from the study of special epitopes in humoral responses to the characterization of cancer biomarkers and antibody characteristics to the characterization of protein-protein interactions and kinase substrates.
